M.tuberculosis strains were grown in Middlebrook 7H9 broth supplemented Selleckchem Screening Library with OADC (Oleic acid, Bovine albumin fraction V, dextrose-catalase) from Difco laboratories, USA and 0.05% Tween 80 (Sigma). M.smegmatis was grown either in Middlebrook 7H9 supplemented with glycerol or on Middlebrook 7H11 plates. Transformation in Escherichia coli DH5α was carried out using heat shock method [14] and in M. smegmatis mc 2 155 by electroporation [19] using Gene Pulser (Bio Rad Laboratories Inc.
